And what mean standard tune?

Not sure I understand what you're asking...

"You Are My Sanity" is in standard (EADGBE) tuning.

I don't understand where you're getting C# from. The song is in D major, or B minor.

Alright, I see what you're saying now. But I disagree. Tim plays a C# in every section of the structured song, and it shows up all over the improv. Listen for it.
